Patent No: 6,509,156 titled ‘DNA cloning method relying on the E. coli recE/recT recombination system’ with the abstract: The invention relates to methods for cloning DNA molecules using recE/recT-mediated homologous recombination mechanism between at least two DNA molecules where one DNA molecule is a circular or linear DNA molecule and the second DNA molecule is a circular DNA molecule, and the second DNA molecule contains two regions with sequence homology to the first DNA molecule. Competent cells and vectors are also described.

Patent No: 4,356,270 titled ‘Recombinant DNA cloning vehicle’ with the abstract :The Specification discloses: 1. Recombinant microbial cloning vehicles comprising heterologous DNA coding for the expression of mammalian hormone (e.g., somatostatin) and other polypeptides, including plasmids suited for the transformation of bacterial hosts. The latter incorporate a regulon homologous to the host in its untransformed state, in reading phase with the structural gene for the heterologous DNA; 2. Cloning vehicles coding for the microbial expression of a protein variously comprising (a) a polypeptide hapten and additional protein sufficient in size to confer immunogenicity on the product of expression, which may find use in raising antibodies to the hapten for assay use or in the manufacture of vaccines; and (b) a desired polypeptide product and additional protein from which the desired product may be cleaved; and 3. Methods of preparing synthetic structural genes coding for the expression of mammalian polypeptides in microbial cloning systems.

Patent No: 4,590,163 titled ‘Broad host range DNA cloning system for gram-negative bacteria’ with the abstract: Novel compositions and methods for cloning DNA in a broad range of Gram-negative bacteria. Relatively small vectors are prepared substantially free of DNA sequences coding for transfer functions, while having a broad host range replication function. While the plasmid is not self-transmissible, it is transmissible by means of a helper plasmid, which desirably has a narrow host range replication function. Exogenous DNA may be inserted into the first plasmid and conjugally transferred to the target host by means of a convenient bacterial host.

Patent No: 4,321,365 titled ‘Oligonucleotides useful as adaptors in DNA cloning, adapted DNA molecules, and methods of preparing adaptors and adapted molecules’ with the abstract : Synthetic oligonucleotides have been designed and prepared which are useful in the molecular cloning of a variety of DNA molecules. By means of such oligonucleotides, genetic informational material, e.g., DNA can be joined to a cloning vehicle and transferred into host cells by transformation. Additionally, a method for determining whether genetic informational material has been transferred into transformed host cells has been developed.
